2.2 Features
The CellTramAir, CellTramOil and CellTramvario are manual piston pumps. Using a movable
piston, the CellTram generates differences in pressure which are transferred directly to a capillary
via a pressure tube. The piston is moved in the CellTram using the dial. A clockwise rotation, i.e.
to the right, results in an increase in pressure, the capillary contents are pressed out. A
counterclockwise rotation, i.e. to the left, results in a decrease in pressure, aspiration into the
capillary takes place. To assist fine pressure adjustment, the dial has a scale.
The capillary is inserted into a capillary holder which in turn provides the connection to the
pressure tube. The universal capillary holder enables permanent mounting of various capillaries
in the laboratory.
The CellTramAir, CellTramOil and CellTramvario have a very ergonomic design which enables
easy handling, particularly in conjunction with all control units of Eppendorf micromanipulators.
2.2.1CellTramAir
The CellTramAir uses air for pressure transmission. This guarantees clean and simple handling
of the piston pump.
A complete revolution of the dial corresponds to a change in volume of approx. 88L.
The position of the piston is displayed in the piston window on a scale from 0 to 15mm
corresponding to the strokes.
2.2.2CellTramOil/CellTramvario
The CellTramOil and CellTramvario use oil for pressure transmission which, unlike air, can
hardly be compressed. This ensures its operating cycles are very controlled and constant. The
simple filling technique reduces the filling time and simplifies oil filling. To ensure controlled,
constant pressure transmission, the cylinder, pressure tube and part of the capillary are filled with
oil. A complete revolution of the dial (coarse drive) corresponds to a change in volume of approx.
9.6L.
The CellTramvario has an additional fine drive dial. The transmission ratio between fine drive
and coarse drive is 10:1. A revolution of the fine drive corresponds to a change in volume of
approx. 0.96L. This enables ultra-fine cell movements.
The position of the piston can be observed in the transparent cylinder. The filling bore with the
plug screw is at the top of the cylinder. The pressure tube is connected to the CellTramOil/
CellTramvario with the aid of the tube connection screw. The pressure tube has a joint at both
ends and thus connects the CellTramOil/CellTramvario and the universal capillary holder.

3.1 User profile
The device and accessories may only be operated by trained and skilled personnel.
Before using the device, read the operating manual carefully and familiarize yourself with the
device's mode of operation.
3.2 Intended use
The CellTram has been designed and manufactured for use in the field of Intracytoplasmic sperm
injection (ICSI) in conjunction with Eppendorf VacuTips and Transfertips. The CellTram is thus
considered a medical device within the scope of Directive 93/42/EEC.
The CellTram is intended exclusively for indoor use, and for operation by qualified staff.
3.3 Warnings for intended use
WARNING! Risk of injury due to flying capillaries and glass splinters.
If exposed to high pressures, capillaries can detach themselves from the grip heads and

Capillaries can crack as a result of incorrect handling.
Wear protective goggles.
Never aim capillaries at people.

Aspirating oil with the syringe may lead to the formation of tiny air bubbles. This should

be avoided as these air bubbles may be transferred to the CellTram, which may impair
or slow down the filling process. If the syringe contains air bubbles, these should be
collected by long-term storage. Large air bubbles can be pressed out during filling.

4.3.1Heating the syringe
1. Screw the filling hose on to the tip.
2. Aspirate at least 3mL of oil into the syringe.
4.3.2Connecting the filling tube
1. Turn the rotary knob to move the piston to
the initial position.
2. Unscrew the blind screw.
3. Screw the filling tube in the filling bore.

Prerequisites
The piston is in the starting position.
1. Hold the CellTram vertically.
2. Push some oil into the piston chamber.
During the first filling, air bubbles may be
present between the piston and the cylinder.

4.3.4Sealing the filling bore
1. Position the CellTram horizontally.
2. Unscrew the filling tube.
3. Fill the filling bore with oil.

4. Screw on the blind screw. When doing so,
make sure that no new air bubbles enter the
bore.
5. Soak up any excess oil using a soft
cellulose cloth.
4.3.5Removing air bubbles
1. If there are still air bubbles in the piston
chamber: slant the CellTram to allow air
bubbles to escape to the filling bore.
Remove the air bubbles.

1. Slightly screw the grip head into the
universal capillary holder.
2. Slide the capillary through the inner O-rings up to the stop into the universal capillary holder.
No stop is felt with capillaries with an inner diameter <0.8mm.

3. Tighten the grip head.
4. In the case of CellTram Oil and CellTram vario, carefully wipe any excess oil off of the
universal capillary holder and the capillary using a soft cellulose cloth if necessary.
When using Eppendorf Femtotips
1. Screw the Femtotip adapter into the universal capillary holder.
2. Screw the Femtotip into the Femtotip adapter.
4.5 Setting the angle of the CellTram
1. Loosen the fastening screw in the tilting joint with an Allen key (4mm).
2. Set the CellTram via the tilting joint.
3. Tighten the fastening screw in the tilting joint with an Allen key (4mm).

5Operation
5.1 Holding and transferringcells
CellTramAir:
Hint!
An overpressure or vacuum can be built up in the capillary using the volume of the
piston chamber (cylinder) and the pressure tube. Therefore, the pressure behavior
varies depending on the position of the piston. If the piston is located, e.g., in position
10to 15, the differences in pressure are greater than, e.g., in position 0 to 5. By
carefully selecting the position of the piston, the sensitivity of the CellTramAir piston
pump can be adapted to your requirements.
1. Fix the universal capillary holder into the tool holder of the Eppendorfmicromanipulator.
2. Determine the correct length of the capillary by moving the universal capillary holder in the
tool holder.
3. Determine the correct rotation of the capillary by turning the front knurled screw.
4. Move the micromanipulator upwards (see Operating Manual of the micromanipulator).
5. Position the petri dish.
6. Move the capillary into the medium and ensure its correct positioning by observing it under a
microscope.
Establishing pressure equilibrium in the capillary
7. You have three options:
• Wait until equilibrium has been established.
• Turn the piston clockwise (i.e. to the right) to establish a counter pressure which
compensates the active capillary force.
• Only CellTramAir: loosen the ventilation screw. Tighten the screw again after pressure
compensation.
8. Move the capillary near to the desired cell.
9. Generate a vacuum by turning the dial counterclockwise (to the left).
In hold mode:

Excessive constant pressure can damage the cell. After the cell has reached the
capillary tip, an optimal constant pressure should be set.
In transfer mode:
Hint!
Ensure that pressure is equilibrated sensitively and evenly during cell aspiration into the
capillary.
10. While the cell moves towards the capillary: it may be necessary to turn the dial clockwise (to
the right) to slightly equilibrate the vacuum generated in order that the cell is not fixed too
firmly to the capillary tip.

5.2 Suctioning liquid
Valid for:

1. Dip the capillary into the liquid that is to be aspirated.
2. Turn the rotary knob counterclockwise to the left (by approx. 2revolutions) to aspirate the
liquid by creating a low pressure.
3. Wait for a short period and then loosen the ventilation screw.
4. Retighten the ventilation screw after pressure compensation.

6.1 Replacing O-rings
If the O-rings on the universal capillary holder or on the grip head are damaged, they can be
replaced.
1. Remove the O-rings and the sealing washer with the extraction tool.
2. Insert the 2 new O-rings and a new sealing washer in the correct order.
3. Make sure that the 2 sealing washers and 2 matching O-rings are correctly arranged in the
grip head.
6.2 Cleaning
NOTICE! Damage from the use of aggressive chemicals.
Do not use any aggressive chemicals on the device or its accessories, such as

strong and weak bases, strong acids, acetone, formaldehyde, halogenated
hydrocarbons or phenol.
If the device becomes contaminated with aggressive chemicals, clean it
immediately using a mild cleaning agent.
Choose the disinfection method which corresponds to the legal regulations and
guidelines in place for your range of application.
If you have any questions on cleaning, disinfection, decontamination, or the cleaning
