1 About this manual
This manual describes the Hydrolysis Unit B-411/E-416 and provides all information required for its safe operation and to maintain it in good working order. It is addressed to laboratory personnel in particular.
NOTE
The symbols pertaining to safety (WARNINGS and ATTENTIONS) are explained in chapter 2.

2 Safety
This chapter points out the safety concept of the instrument and contains general rules of behavior and warnings from hazards concerning the use of the product. The safety of users and personnel can only be ensured if these safety instructions and the safety­related warnings in the individual chapters are strictly observed and followed. Therefore, the manual must always be available to all persons performing the tasks described herein.
2.1 User qualification
The instrument may only be used by laboratory personnel and other persons who on account of training or professional experience have an overview of the dangers which can develop when operating the instrument. Personnel without this training or persons who are currently being trained require careful instruction. The present Operation Manual serves as the basis for this.
2 Safety
2.2 Proper use
The instrument has been designed and built for laboratories. It serves for the hydrolysis of products for fat determination with the use of diluted hydrochloric acid.
2.3 Improper use
Applications not mentioned above are improper. Also, applications, which do not comply with the technical data, are considered improper. The operator bears the sole risk for any damages caused by such improper use.
The following uses are expressly forbidden:
Use of the instrument in rooms which require ex-protected instruments.• Use on samples, which can explode or inflame (example: explosives, etc.) due to shock, friction, • heat or spark formation. Use in overpressure situations.•

2 Safety
2.5 Product safety
The Hydrolysis Unit is designed and built in accordance with current state-of-the-art technology. Nevertheless, risks to users, property, and the environment can arise when the instrument is used carelessly or improperly.
The manufacturer has determined residual dangers emanating from the instrument
if the instrument is operated by insufficiently trained personnel.• if the instrument is not operated according to its proper use.•
Appropriate warnings in this manual serve to make the user alert to these residual dangers.
2.5.1 Instrument-related hazards
Pay attention to the following safety notices:
WARNING
Potentially hot surfaces during operation, especially at the glass parts or due to hot hydrochloric acid.
Always be aware of the risk of being burned.•
WARNING
Potential implosion risk if used with damaged glassware.
Beware of damaged or cracked glass parts.•
2.5.2 Safety measures
Always wear personal protective equipment such as protective eye goggles, protective clothing and gloves when working with the instrument.

2.5.3 Safety elements
The main switch of the instrument serves as safety switch. In case of overload or short circuit it • automatically falls back to the zero position.
2.6 General safety rules
Responsibility of the operator The head of laboratory is responsible for training his personnel.
The operator shall inform the manufacturer without delay of any safety-related incidents which might occur during operation of the instrument. Legal regulations, such as local, state and federal laws applying to the instrument must be strictly followed.
Duty of maintenance and care The operator is responsible for ensuring that the instrument is operated in proper condition only, and
that maintenance, service, and repair jobs are performed with care and on schedule, and by autho­rized personnel only.

Spare parts to be used Use only genuine consumables and genuine spare parts for maintenance to assure good system
performance and reliability. Any modifications to the spare parts used are only allowed with the prior written permission of the manufacturer.
Modifications Modifications to the instrument are only permitted after prior consultation with and with the written
approval of the manufacturer. Modifications and upgrades shall only be carried out by an authorized Buchi technical engineer. The manufacturer will decline any claim resulting from unauthorized modifi­cations.

4 Description of function
This chapter explains the basic principle of the instrument, shows how it is structured and gives a functional description of the assemblies.
For Weibull-Stoldt fat determination in food and animal feed, the product must be hydrolyzed to make the fat accessible for extraction. Thus a reproducible fat determination is possible. The hydrolysis also frees fat-like substances which are mechanically surrounded by different constitu­ents (carbohydrates and protein substances in undamaged cells or starch membranes). This is also true for colloid-disperse fractions (protein) which, as a result of surface forces, coat the fat drops (milk, cream, cheese) and for certain fat fractions which are bound, chemically or adsorptively to other components (phosphatide-protein complexes in yeast, eggs, germs, etc.).

4.3 Principle of hydrolysis
The sample is boiled in diluted hydrochloric acid to break down proteins and high-molecular carbohydrates into acid-soluble con­stituents.

Afterwards the sample is filtered over a prepared glass sample tube and dried. The celite distributes the hydrolysate evenly in the tube, prevents clumping and facilitates the extraction to be carried out.
The fat can now be extracted with an appropriate solvent.

5 Putting into operation
This chapter describes how the instrument is installed and gives instructions on initial startup.
NOTE
Inspect the instrument for damages during unpacking. If necessary, prepare a status report immedia­tely to inform the postal company, railway company or transportation company.
Remove the transportation lock at the bottom of the instrument. Keep the original packaging for future transportation.
5.1 Installation site
Install the instrument on a clean, flat and stable base.
ATTENTION
To ensure the instrument safety do not place any object on top of it. Provide a distance of at least 30 cm between the instrument and another object or the wall. There should be no containers, chemi­cals or other devices located behind the instrument.
5 Putting into operation
We recommend to operate the instrument under a ventilation hood. Do not place the Extraction Unit under the same hood.
5.2 Electrical connections
WARNING
Make sure that the voltage on the socket corresponds to the voltage given on the type plate of the instrument.
Always connect the instrument to an earthed socket. External connections and extension cables must be provided with an earthed conductor lead (3-pole couplings, cable or plug equipment) as the mains lead has a molded plug, thus avoiding risks due to inadvertent defective wiring.
Make sure that no electric sparks form in the instrument or its surroundings as they might damage the instrument.
Connect the Hydrolysis Unit B-411/E-416 to the mains with the power cord contained in the scope of delivery.
5.3 Connecting the suction hose to the suction tube and the pump
The suction hose is connected to the hose connection (Ø 10 mm) on the suction tube. The fumes which result from hydrolysis and the wash water resulting from filtering the sample are extracted with the pump. The suction tube can be connected to a water jet pump or to a vacuum pump with an intermediate vessel. In order to achieve efficient filtration, the used pump should have a good suction power and should be resistant to the chemicals used.

5.4 Installing the rubber couplings
The rubber couplings can be carefully installed on the suction tube and the sample aspiration tube after having moistened them with water. During the installation, make sure that the collar of the glass part abuts on the receptacle of the rubber part. The receptacles for the glass sample tubes are provided with an antistick coating, so that they can be easily installed and disassembled. In order to avoid broken glass during the installation of the rubber couplings, the glass part must be held as near as possible to the receptacle collar.

5.5 Filtration positioning
The distance between the sample aspiration tube and the digestion vessel must be approx. 5 mm. In order to guarantee optimal filtration, it is necessary to correctly position the cap.

6 Operation
This chapter gives examples of typical instrument applications and instructions on how to operate the instrument properly and safely.
WARNING
Risk of injury.
Never operate the instrument when glassware is damaged.•
ATTENTION
Hot, caustic hydrochloric acid is used for the operation of the instrument. For this reason, personal protective equipment must be worn at all times when using this instrument or working in its imme­diate proximity.
6.1 Sample weight
6 Operation
The sample weight depends on the fat content of the sample. It should be chosen in a way that the final weight is between 0.5 and 1.5 g of fat. The maximum sample weigth is 10 g.
6.2 Preheating
The “preheating” position serves to preheat the instrument. This preheating procedure takes about 10 minutes. Thus an initial boiling point of the sample is achieved in a shorter period of time. The preheating must always be carried out before a hydrolysis process is started.
6.3 Preparing the digestion vessels
Put 5 g celite 545 into the digestion vessel.

Add 50 ml HCl, 4 mol / l and shake the tube to mix the sample with the celite and the HCl.
Add another 50 ml HCl, 4 mol / l by pouring it along the side walls of the digestion vessel to remove probable residues from the tube wall.
In case of liquid samples, the acid concentration should be adjusted to the water content of the sample.

6.4 Preparing the glass sample tubes
Evenly distribute approx. 50 g quartz sand (grain size 0.3 - 0.9 mm). The grain size is important, as in case it is too small, it will clog the glass sample tube and in case it is too large, the celite will pass the glass sample tube during the filtration step.

6.5 Choosing the heat output
When the preheating procedure is finished, the hydrolysis can be started. For this purpose we recom­mend to switch the energy regulator to position 2 for four samples and to position 3 for six samples. Depending on the boiling behavior, less heat can be provided when setting the regulator to position 1 or more heat can be provided by setting it to position 3.
6.6 Hydrolysis
To carry out a hydrolysis proceed as follows:
Reduce the heat output (position 2 - 3)• Insert the vessels containing the samples into the instrument• Put the prepared glass sample tubes into the rubber couplings• Close unused positions with the supplied stoppers Ø 45 mm on the suction tube• Put the caps onto the digestion vessels and install the sample aspiration tube• As soon as the sample starts boiling switch on the waterjet pump.• The hydrolysis time depends on the sample. From the start of the boiling on it takes a minimum of • 15 and a maximum of 60 minutes (usually 30 minutes is an appropriate time).
6 Operation
Foaming can be suppressed by the addition of small acid injections (HCl 4 mol/l). It is recommended to switch on the external pump only after the samples are boiling evenly and no further foam is to be expected. Thus a premature sample extraction is avoided.

After expiration of the hydrolysis time, switch off the heating and add hot distilled water (40 - 50 °C) to each digestion vessel to a level of approx. 1 cm under the sample aspiration tube.
To filter the hydrolysate solution over the glass sample tubes, raise the lift device to immerse the sample aspiration tubes into the solu­tion. As soon as the contents of the individual digestion vessels are completely transferred, rinse them with hot distilled water (40 - 50 °C) portion by portion. The rinse volume until neutral reaction is approx. 250 ml per sample.

6.8 Drying the hydrolyzed samples
Before drying the hydrolyzed samples stir the sample layer and the celite layer (without touching the sand layer) with a spatula to loosen the pulp. Afterwards wipe the spatula with fat free cotton wool or a tissue and put the tissue into the sample.
Dry the glass sample tubes in a vacuum oven ( 4 h at 100°C/200mbar), in an oven ( 8 h at 100°C) or in a microwave oven. Using a microwave oven makes the drying much faster. However, it is more awkward due to the fact that the temperature in the sample can easily become too high (> 105 °C) if an inappropriate heating power has been chosen. Therefore, we suggest the following:
Step: 18 minutes with an output of 640 W1. Step: 10 minutes with an output of 480 W for 4 samples 2.
13 minutes with an output of 480 W for 6 samples
The optimal parameters for the microwave may depend on the model.
The sample is dry when there is no moisture to be seen on the glass wall.
6 Operation
ATTENTION
The plastic holders for the glass sample tubes and paper thimbles for the E-812/E-816 are not microwavable.
6.9 Preparing the extraction
Fill the dried, cooled, glass sample tubes with 20 g of quartz sand (grain size 0.3-0.9 mm). They are now ready for extraction. The glass sample tubes are optimized for use either with the Extraction System B-811 or with the Extraction Unit E-812/816.

7 Maintenance
This chapter gives instructions on all maintenance work to be performed in order to keep the instru­ment in good working condition.
WARNING
All maintenance and repair work requiring the opening or removal of instrument covers may only be carried out by trained personnel and with the tools provided for this purpose.

Prior to all maintenance work on the instrument switch off the power supply and remove all • sources of flammable vapor.
ATTENTION
Always wear personal protective equipment such as protective eye goggles, protective clothing and gloves when maintaining the instrument.
7 Maintenance and repairs
7.1 Housing
Check the housing for defects (switches, plugs) and clean it regularly with a damp cloth and common chromium steel cleaners.
ATTENTION
Never use organic solvents (except for ethanol) as cleaning agents as these might damage the instru­ment.
As the housing is made of stainless steel and plastic, any acid drops must be wiped away imme-• diately with a damp cloth.
7.2 Heating chamber
ATTENTION
The surface of the heating chamber can get very hot. Always let the instrument cool down sufficiently before carrying out any maintenance work at the heating chamber.
Remove the upper insulation plate a above the heating chamber. You can now clean the insulation with a moist cloth or a vacuum cleaner.

7.3 Glass parts
The glass parts can be taken out and cleaned with commercially available cleaning agents or in an ultrasonic bath. After the glass parts have been cleaned and fully dried, check each part visually for cracks, scratches and for any parts or sections that might have splintered off. Take out and replace any damaged glass parts.
7.4 Rubber couplings
Rinse the rubber couplings regularly with water.
7.5 Functional test
A functional inspection should be carried out annually or in case of weak performance of the instru­ment. To carry it out, proceed as follows:
Clean the instrument as described above in this chapter.• Visually inspect the instrument, the lift device, the connecting cable, the glass parts and the hose • connections. Start-up the instrument with the energy regulator in the preheating position and check, whether a • preheating is done. In the case of technical problems, contact a Buchi customer service office.•

Pull out the fuse holder • a Replace the defective mains fuse with a new • one with same performance:
- 230V: T 5A L250V•
- 120V: T10A L250V• Reinsert the fuse holder•

8 Shutdown, storage, transport and disposal
8 Shutdown, storage, transport and disposal
This chapter instructs how to shut down the instrument, how to pack it for storage or transport, and specifies the storage and shipping conditions.
8.1 Storage and transport
WARNING
Unplug the mains cable and remove and thoroughly rinse the suction tube. Clean the instrument thoroughly, remove all chemical residues and wash the glass parts.
Store and transport the instrument in its original packaging.
8.2 Disposal
To dispose of the instrument in an environmentally friendly manner, a list of materials is given in chapter 3. This helps to ensure that the components are separated and recycled correctly. Make especially sure to dispose of the gas springs appropriately. Please follow valid regional and local laws concerning disposal.

Always state the product designation and the part number when ordering spare parts.
