CRT® bacteria

Instructions for Use

Description

CRT bacteria is used to determine the mutans streptococci
and lactobacilli count in saliva by means of selective
culture media.

Indication

• In vitro diagnostics

• Bright agar surface: determination of lactobacilli count
in saliva

• Blue agar surface: determination of mutans streptococci
count in saliva, or plaque

Contraindication

Do not use CRT bacteria during treatment with antibiotics.
After such treatment, at least two weeks should pass
before CRT bacteria is used. After the use of an
antibacterial mouthrinse, at least 12 hours should
pass before CRT bacteria is used.

Application

The test is professionally conducted by the dentist and
skilled personnel.

Step by step procedure

1. Have the patient stimulate salivation by chewing the

enclosed paraffin pellet.

2. Collect the saliva in a suitable container.

Tip: Take this opportunity to measure also the
salivation rate and the buffer capacity of saliva
with CRT buffer from Ivoclar Vivadent.

3. Remove the agar carrier from the test vial.

4. Place a NaHCO3-tablet at the bottom of the vial.

5. Carefully remove the protective foils from the two agar

surfaces. Do not touch the agar.

6. Thoroughly wet both agar surfaces with saliva,

using a pipette. Avoid scratching the agar surface.
Tip: Hold the agar carrier slightly oblique.

7. Allow excess saliva to drip off.

8. Slide the agar carrier back into the vial and close the
vial tightly.

9. Use a waterproof pen to note the name of the patient
and the date on the vial.

10. Place the test vial upright in the incubator, e.g.
Cultura/Ivoclar Vivadent, and incubate at 37 °C / 99 °F
for 48 hours.

11. After removal of the vial from the incubator, compare
the density of the mutans streptococci and lactobacilli
colonies with the corresponding evaluation pictures in
the enclosed model chart.
Tip: Hold the agar carrier slightly oblique under a light
source to facilitate the evaluation.

Modified procedures

A slight modification of the test procedure permits the
determination of the mutans streptococci count in plaque:

• Remove plaque with a fine brush.

• Apply plaque on the blue agar without scratching the

surface. Up to four samples may be applied on the same
carrier in a parallel pattern without any difficulty.

Note: Apply a small amount of water to the NaHCO

tablet in case not enough saliva drips onto the tablet.

The preparation of sampling and incubation are carried out
according to the step-by-step procedure described in these
Instructions. As far as the evaluation is concerned, this test
only determines whether or not mutans streptococci are
present.

Evaluation

Findings of 10

CFU or more of lactobacilli and mutans

streptococci per ml saliva indicate a high caries risk.

Note

Leaving the test vial in the incubator for an additional day
or two does not influence the number of CFUs.

Disposal

• Do not use new agar carriers that appear to be covered
with microorganisms.

• Before disposing of used agar carriers, disinfect them
with a suitable agent or autoclave them in an
autoclaving bag.

Storage

• Store CRT bacteria in the refrigerator
(2–8 °C / 36–46 °F)

• Do not freeze the material

• Avoid temperature fluctuations

• Protect from light

• Shelf life: see date of expiration

• Do not use CRT bacteria after the indicated date of
expiration

Gebrauchsinformation

Beschreibung

CRT bacteria dient zur Bestimmung der Anzahl der Mutans
Streptokokken und der Laktobazillen im Speichel mit Hilfe
jeweils selektiver Nährböden.

Indikation

• In vitro Diagnostik

• Helle Agarfläche: Bestimmung der Laktobazillen im
Speichel

• Blaue Agarfläche: Bestimmung der Mutans
Streptokokken im Speichel bzw. in der Plaque

Kontraindikation

CRT bacteria nicht während einer Antibiotikabehandlung
verwenden. Eine entsprechende Therapie sollte mindestens
14 Tage zurückliegen. Die Verwendung einer antibakteriellen
Mundspüllösung sollte 12 Stunden zurückliegen.

Anwendung

Die Anwendung erfolgt professionell durch Zahnarzt,
Dentalhygienikerin oder Prophylaxehelferin.

Step by step

1. Patient: Speichelfluss durch Kauen des beiliegenden

Paraffinpellets stimulieren.

2. Speichel in einem geeigneten Gefäss sammeln.

Tip: Hier die Bestimmung der Speichelfliessrate
und die Bewertung der Pufferkapazität des
Speichels mit Hilfe von CRT buffer/Ivoclar Vivadent
gleich mitberücksichtigen.

3. Den Agarträger aus dem Probenröhrchen entnehmen.

4. Eine NaHCO3-Tablette auf den Boden des Röhrchens

geben.

5. Schutzfolien von beiden Agarflächen vorsichtig
abziehen; Agarflächen nicht berühren.

6. Beide Agarflächen mit Hilfe einer Pipette vollständig
mit Speichel benetzen, ohne diese zu zerkratzen.
Tip: Agarträger leicht schräg halten.

7. Überschüssigen Speichel abtropfen lassen.

8. Den Agarträger ins Röhrchen zurückgeben und fest
verschliessen.

9. Mit einem wasserfesten Stift das Röhrchen mit dem
Namen des Patienten und dem Datum kennzeichnen.

10. Das Röhrchen 48 Stunden bei 37 °C im Brutschrank,
z.B. Cultura/Ivoclar Vivadent, aufrechtstehend
aufbewahren.

11. Nach Entnahme des Röhrchens aus dem Brutschrank
die Koloniendichte der Mutans Streptokokken bzw.
Laktobazillen jeweils mit den entsprechenden
Abbildungen der beiliegenden Model Chart vergleichen.
Tip: Um das Auswerten zu erleichtern, Agarträger
schräg unter eine Lichtquelle halten.

Modifikation

Eine Modifikation des Testverfahrens ermöglicht die
Erfassung der Mutans Streptokokken in der Plaque:

• Mit einem feinem Pinsel Plaque entnehmen.

• Plaque auf den blauen Agar ohne diesen zu zerkratzen

abstreichen; bis zu vier Proben können problemlos
parallel aufgetragen werden.

Hinweis: Auf die NaHCO

-Tablette etwas Wasser geben,

da eventuell nicht genug Speichel darauf tropft.

Probenvorbereitung und Inkubation verlaufen analog der
Step by step Anleitung. Hinsichtlich der Auswertung ergibt
sich die Information Mutans Streptokokken ja bzw. nein.

Auswertung

Ein Befund ab 10

KBE Laktobazillen bzw. Mutans Strepto-

kokken pro ml Speichel deutet auf ein hohes Kariesrisiko.

Besondere Hinweise

Steht das Probenröhrchen ein oder zwei Tage länger im
Brutschrank, so wirkt sich dies nicht auf die Kolonienzahl aus.

Entsorgung

• Unbenutzte Agarträger mit Keimbewuchs nicht
verwenden, sondern entsorgen.

• Gebrauchte Agarträger vor dem Wegwerfen mit einem
geeigneten Desinfektionsmittel benetzen oder in einem
Autoklavierbeutel verpackt autoklavieren.

Mode d'emploi

Description

Le CRT bacteria sert à évaluer le nombre de Streptocoques
mutans et de lactobacilles dans la salive. La méthode est
basée sur l'utilisation d'une solution nutritive sélective.

Indications

• Diagnostic in vitro

• Bande de gélose claire: évaluation du nombre de
lactobacilles dans la salive

• Bande de gélose bleue: évaluation du nombre de
Streptocoques mutans dans la salive ou dans la plaque
dentaire

Contre-indications

Ne pas utiliser le CRT bacteria pendant un traitement
antibiotique. Respecter un délai de 14 jours après un
traitement antibiotique avant d'utiliser CRT bacteria.
De même, un délai de 12 h est à respecter quand on
utilise un bain de bouche antibactérien.

Réalisation

Le test doit être réalisé par le chirurgien-dentiste,
l'hygiéniste dentaire ou l'assistante en prophylaxie.

Procédure étape par étape

1. Stimuler la sécrétion salivaire du patient en lui donnant

la tablette de paraffine ci-jointe à mâcher.

2. Recueillir la salive dans un gobelet gradué.

Remarque: Le CRT buffer de Ivoclar Vivadent permet à
la fois d'évaluer le taux de sécrétion salivaire et le
pouvoir tampon de la salive.

3. Ôter le support de gélose du tube de test.

4. Mettre un cachet de NaHCO3sur la base du tube de

test.

5. Ôter délicatement les feuilles de protection des deux
surfaces des supports de gélose ; Ne pas toucher les
surfaces des supports de gélose.

6. Humecter complètement de salive les deux surfaces du
support de gélose à l'aide d'une pipette, sans toucher
la gélose.
Remarque: Pencher légèrement les supports de gélose.

7. Laisser égoutter l'excès de salive.

8. Remettre le support de gélose dans le tube de test et
bien refermer ce dernier.

9. A l'aide d'un stylo indélibile, indiquer le nom du patient
et la date du prélèvement sur le tube de test.

10. Conserver le tube de test en position verticale
dans l'incubateur, par ex. l'incubateur Cultura de
Ivoclar Vivadent, pendant 48 heures à 37 °C.

11. Après avoir sorti le tube de test de l'incubateur,
comparer la densité des colonies de Streptocoques
mutans ou de lactobacilles avec la représentation
correspondante de la fiche d'interprétation jointe.

Modification

Une modification de la procédure de test permet le
recensement des Streptocoques mutans dans la plaque
dentaire:

• Prélever de la plaque dentaire avec un pinceau fin.

• Poser la plaque sur le support de gélose bleu en faisant

attention à ne pas griffer ce dernier. Il est possible de
disposer jusqu'à 4 échantillons en même temps sur le
même support de gélose.

Remarque: Humecter le comprimé de NaHCO

avec
quelques gouttes d'eau dans le cas où la viscosité de la
salive ne permet pas d'imprégner ce comprimé.

La préparation des échantillons et l'incubation sont les
mêmes que celles décrites dans la procédure étape par
étape ci-dessus. En ce qui concerne l'interprétation, ce test
démontre si oui ou non il y a présence de Streptocoques
mutans.

Evaluation

Un résultat supérieur à 10

bactéries lactobacilles et
Streptocoques mutans par ml de salive indique un risque
de carie élevé.

Remarque particulière

Si le tube de test est laissé dans l'incubateur un ou deux
jours de plus, cela n'affecte pas le nombre de colonies.

Elimination

• Ne plus utiliser un support de gélose recouvert de

germes.

• Humecter le support de gélose utilisé avec un

désinfectant adéquat ou le stériliser à l'autoclave dans
un sachet de stérilisation avant de le jeter.

Recommandations de stockage

• Conserver CRT bacteria dans un réfrigérateur (2–8 °C).

• Ne pas angeler

• Eviter les variations de température.

• Garder le produit à l'abri de la lumière.

• Conservation: voir la date de péremption.

• Ne pas utiliser CRT bacteria après la date de

péremption.

Istruzioni d’uso

Descrizione

CRT bacteria determina il numero di Streptococco mutans e
di Lactobacilli presenti nella saliva mediante un terreno di
Agar selettivo.

Indicazioni

• Diagnosi in vitro

• Lato chiaro del terreno: determinazione dei Lactobacilli
presenti nella saliva

• Lato scuro del terreno: determinazione dello
Streptococco mutans presente nella saliva o nella
placca.

Controindicazioni

Non utilizzare CRT bacteria durante una cura antibiotica.
Enecessario lasciar trascorrere almeno 14 giorni dalla
conclusione della cura prima di effettuare il test. Non
utilizzare colluttori antibatterici nelle 12 ore antecedenti
il test .

Esecuzione

Il test viene eseguito ambulatorialmente dallodontoiatra,
dalligienista dentale o dallassistente alla poltrona.

Step by Step

1. Paziente: stimolare la salivazione masticando la

pasticca di paraffina presente nella confezione.

2. Raccogliere la saliva in unapposito misurino.

Consiglio: a questo punto può essere valutato il grado
di flusso salivare e può essere eseguito il test CRT buffer
per la determinazione del potere tampone della saliva.

3. Estrarre dalla provetta il terreno di Agar.

4. Applicare una pasticca di NaHCO3- alla base della

provetta.

5. Staccare con attenzione la pellicola protettiva del
terreno di Agar: non toccare il terreno di coltura.

6. Utilizzando la saliva precedentemente raccolta,
umettare entrambi i lati del terreno di Agar, senza
strisciarli, con lausilio di una pipetta.
Consiglio: tenere lasta leggermente inclinata in modo
da far scorrere agevolmente la saliva sulla superficie.

7. Eliminare gli eventuali eccessi di saliva.

8. Inserire lasta del terreno di Agar nella provetta e
chiudere bene.

9. Con una penna ad inchiostro indelebile, contrassegnare
la fiala con il nome del paziente e la data.

10. Inserire la provetta in posizione verticale in un
incubatore, p. es.: Cultura/Ivoclar Vivadent, e lasciarla
maturare per 48 ore a 37 °C.

11. Prelevare la provetta dallincubatore e valutare la
presenza delle colonie di Lactobacilli e Streptococco
mutans confrontando il terreno di Agar con la Model
Chart allegata.
Consiglio: per agevolare la valutazione, tenere lasta
del terreno di Agar leggermente inclinata sotto una
fonte di luce.

Alternativa d utilizzo

Un utilizzo alternativo del test consente di rilevare la
presenza di Streptococco mutans nella placca.

• Prelevare un campione di placca con una pinzetta.

• Depositare il prelievo di placca direttamente sul terreno

di Agar blu scuro senza danneggiarlo. Ben posizionati
sul terreno di Agar, possono essere confrontati fino a
4 campioni di placca contempora-neamente.

Avvertenza: Aggiungere un po' d'acqua alla complessa
di NaHCO

qualora non si riesca a far gocciolare un

quantitativo sufficiente di saliva sulla pastiglia.

Preparazione ed incubazione si svolgono come descritto
nella fase di esecuzione del test precedente. Nel caso della
placca non viene fatta una valutazione quantitativa ma
solo un rilevazione “si” o “no” della presenza di
Streptococco mutans nella placca.
