HOW TO START THE SYSTEM

Switch on instrument, computer and touch screen

If necessary, open the easyMAG software using desktop shortcut or go to Start/Program/easyMAG and select easyMAG software

Wait until status led turns on either green or red fixed state

Enter login name and password with appropriate daily use rights

HOW TO DEFINE EXTRACTION REQUEST

Define extraction request
1. Define Matrix, Protocol, input Volume (mL), Eluate volume (µL), type of sample (lysed or non­lysed=Primary)
2. Then enter Sample ID (manually or using barcode)
3. Enter lysis buffer lot number (Optional) in case of lysed sample
4. Select:
To save request or click ENTER
2 options to define extraction request(s)
To define a new request
or
To auto-generate incremented requests in the range [1…120]

List size from 1 to 120 requests

1. Create new run
2. Enter run name
3. Define lysis workflow
On-board or Off-board
4. Define silica incubation workflow

1. Select appropriate run
2. Select sample(s) ID you want to

Add primary sample to vessel
On the instrument
1. Load sample vessel on the instrument
2. Scan position then sample vessel barcode number
3. Finally add primary sample*
*If Sample Guidance ON, identify sample:

Perfom lysis on board
1. Start Dispense Lysis
2. During 10 min lysis incubation, prepare pre-mix as described in following chapter: “How to prepare pre-mix”

Prepare pre-mix using automatic NucliSENS easyMAG® multi-pipet
1. Dilute Internal Control in diluent
using program 1
2. Add NucliSENS easyMAG® Silica using program 1
3. Dispense premix into micro well strip using program 2
HOW TO PREPARE SAMPLE VESSEL

For lysed sample on-board:
Load aspirator and go to section O ONLY when lysis is finished
and instrument status is idle

For lysed sample off board, load disposables and samples
1. Load aspirator
2. Load sample vessel
3. Scan position then sample vessel barcode number (Mandatory)
4. Add lysed sample to vessel*
*If Sample Guidance ON identify sample:

Add prepared pre-mix to sample vessel and mix using program 3 of the NucliSENS easyMAG® multi-pipet
