40 g des Mediums werden in 1 l destilliertem Wasser suspendiert. Zum Lösen unter Rühren er hit zen. In 200 ml Aliquots abfüllen und für 15 Minuten im Autoklaven bei 121°C sterilisieren. Auf ca. 50 °C abkühlen lassen und in Petrischalen gießen, so dass eine etwa 5 mm dicke Agarschicht entsteht. Erstarren lassen. Zweischichtige Agarplatten zum Schnelltest auf E. coli werden hergestellt indem man eine dünne Schicht (ca. 1 mm) TSA-Agar über fertige TBA-Platten (Best. Nr. CP69.1) gießt. Für jede Untersuchung frisch herstellen.
EINSATZGEBIET
Trypton-Soja-Agar wird vorgegeben von der Europäischen ISO Norm 9308-1:2000 zur Untersuchung von Wasserproben auf die Anwesenheit von coliformen Bakterien. Nach Membranfiltration und Kultur der Proben auf Lactose-TTC-Agar mit Natrumheptadecylsulfat (Best. Nr. CP65.1) werden Subkulturen aller gelben bis orangefarbenen Kolonien mit gelbem Hof auf Trypton-Soja-Agar angelegt. Nach Bebrütung werden die Kolonien durch Oxidasetest auf einem Filterpapier analysiert. Oxidase-negative Kolonien werden als coliforme Bakterien gezählt. Zum Schnelltest auf E. coli wird die filtrierte Membran direkt auf der TSA­Agarplatte oder auf einer zweischichtigen TSA/TBA Kombinationsplatte inkubiert. Coliforme Bakterien sind Gram-negative, stäbchenförmige Bakterien, die keine taxonomische Gruppe darstellen, sondern durch die Eigenschaft charakterisiert sind, aus Lactose unter Säure und Gasbildung Energie gewinnen zu können. Sie kommen vor allem im Darm von Warmblütern vor, können allerdings auch einige Zeit außerhalb der natürlichen Umgebung überleben und sind damit bestens geeignet zur Untersuchung von Wasserproben auf fäkale Verunreinigungen. Allerdings leben einige coliforme Bakterien auch im Boden und in Oberflächenwassern, so dass eine entsprechende Verunreinigung von Wasserproben auch auf andere Fehler in der Wasseraufbereitung zurückzuführen sein kann. Die Europäische ISO Norm 9308-1:2000 legt ein Referenzverfahren fest für den Nachweis und die Zählung von coliformen Bakterien in jedem Wasser für den menschlichen Gebrauch. Vorgegeben werden zwei Verfahren – ein Standardtest, der besonders für keimarmes Wasser geeignet ist und ein Schnellverfahren zur kurzfristigen, aber weniger sensitiven Analyse.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Agar bei einer Temperatur von 36 ± 2 °C für 18-24 Stunden.

PREPARATION (EN ISO 9308-1:2000 – B.3)
Suspend 40 g of the medium in 1 l distilled water. Solubilise under heating with frequent agitation. Pour aliquots of 200 ml each and sterilise for 15 minutes at 121 °C in an autoclave. Let cool to approx. 50 °C and pour into Petri dishes (at approx. 5 mm thickness). Let cool. Double layer agar plates for speedy testing of E. coli are produced by pouring a thin layer (approx. 1 mm) of TSA-agar over ready-to-use TBA-plates (Art. No. CP69.1). Prepare freshly for each test.
USES
Tryptone Soy Agar is prescribed by European ISO Standard 9308-1:2000 for analyzing water samples to determine the presence of coliform bacteria. After filtering the membranes and culturing the samples on TTC-Chapman-Agar with Sodium Heptadecylsulphate (Art. No CP65.1), subculture all yellow to orange­coloured colonies with a yellow corona on Tryptone Soy Agar. Analyze the colonies with an oxidase test on filter paper after incubation. Oxidase-negative colonies should be counted as coliform bacteria. For a quick E. coli test, place the filtered membrane directly on the TSA-agar plates or on a double layer TSA/TBA combination plate. Coliform bacteria are Gram negative, rod-shaped bacteria which do not represent a taxonomic group, but are characterized for being able to produce energy from lactose under acid and aerosis. They are prevalent particularly in the intestines of warm-blooded animals. They can, however, survive for a time outside their natural environment and are therefore extremely suitable for analyzing water samples for faecal contamination. However, some types of coliform bacteria do in fact live in the soil and in surface waters with the result that the respective contamination of the water samples may be ascribed to other errors during treatment of water. European ISO Standard 9308-1:2000 defines reference procedures for the detection and enumeration of coliform bacteria in all types of water meant for human use. Two procedures are given – a standard test which is especially suitable for water low in germs and a quick-test for short term but less sensitive analysis.
