HERSTELLUNG
111 g des Mediums werden in einem Liter destillierten Wasser suspendiert. Man mische gut und erhitze unter häufigem Rühren/Schütteln, bis das Medium vollständig gelöst ist und lasse eine Minute lang kochen. Man sterilisiere 15 Minuten lang bei 121 °C und gieße in Petrischalen.
EINSATZGEBIET
Mannit-Kochsalz-Agar ist ein Medium, das nach den Angaben von Chapman zur Isolation von mutmaßlich pathogenen Staphylokokken (Chapman, 1945, J. Bact. 50:201-3). Es stellt ein hochselektives Medium dar, auf dem auch große Probenmengen ausgestrichen werden können. Die hohe Konzentration an Natriumchlorid in dieser Formulierung inhibiert das Wachstum der meisten anderen Bakterien. Durch den Abbau von Mannitol durch Mannitol-fermentierende pathogene Staphylokokken wird das Medium sauer und wechselt die Farbe von Pink nach Gelb. Mannitol­fermentierende pathogene Staphylokokken wachsen also in meist großen Kolonien, die von einem gelben Hof umgeben sind. Kolonien nicht-pathogener Staphylokokken wachsen als meist kleine Kolonien, die von einem roten oder purpurfarbenen Hof umgeben sind. Zur Bestätigung dieser Kolonien als pathogener Staphylococcus durch die Detektion der Lipase-Aktivität empfehlen wir die Zugabe von 5 % Eigelbemulsion (Best.-Nr. 0402.1) zum Agar. Die Lipaseaktivität resultiert in einem gelb-opaken Hof um die Kolonien von Lipase-produzierendem Staphylococcus.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Medium / Agar bei einer Temperatur von 30-35 °C für 18 - 72 Stunden.

PREPARATION
Suspend 111 g of the medium in one liter of distilled or deionized water. Mix well. Heat with frequent agitation until complete dissolution. Boil for one minute and sterilize at 121 ºC for 15 minutes. Pour intro Petri dishes.
USES
Mannitol Salt Agar is a medium prepared according to the recommendations of Chapman for the isolation of presumptive pathogenic Staphylococci (Chapman, 1945, J. Bact. 50:201-3). It is a highly selective medium, onto which large samples may be streaked without danger of overgrowth. The high concentration of sodium chloride included in this formulation inhibits growth of most of the other bacteria. Through degradation of mannitol by the mannitol fermenting pathogenic staphylococci, the medium is acidified and changes in colour from pink to yellow. Thus, mannitol fermenting pathogenic staphylococci grow in mostly large colonies surrounded by a yellow zone, colonies of non-pathogenic staphylococci appear as mostly small colonies surrounded by a red or purple zone. In order to confirm the colonies as pathogenic Staphylococcus through detection of lipase activity, we recommend addition of 5 % egg yolk emulsion (Art. No. 0402.1). Lipase activity results in a yellow opaque zone around the colonies of staphylococci producing this enzyme.
