KARTOFFEL-EXTRAKT-GLUCOSE AGAR
Empfohlen nach der Harmonisierten Methode (Ph. Eur. 6.0), Ph. Eur. 6.3 und 7.0
Zur Kultivierung und Identifizierung von Hefen und Schimmelpilzen
Ph. Eur.

Zusammensetzung in g/l:
Kartoffel-Infus ..................... 4,0
Glucose ............................. 20,0
Agar .................................. 15,0
pH .............................. 5,6 ± 0,2
HERSTELLUNG
MIKROBIOLOGISCHE TESTS

POTATO-EXTRACT-GLUCOSE AGAR
Recommended by the Harmonized Method (Ph. Eur. 6.0) , Ph. Eur. 6.3, and 7.0
For the cultutivation and identification of yeasts and molds
Ph. Eur.
X931
Formulation in g/l:
Potato Infusion ...................... 4.0
Dextrose ............................. 20.0
Agar .................................... 15.0
pH .............................. 5.6 ± 0.2
PREPARATION
Suspend 39 g of the medium in one liter of distilled or deionized water. Mix well and heat with frequent
agitation. Boil for one minute. Sterilize at 118 C for 15 minutes. Cool to 40-45 C and pour into Petri
dishes. The medium is amber and slightly opalescent due to the potato infusion.
USES
Potato-Extract-Glucose Agar is used for the isolation, enumeration and identification of yeast and molds.
This medium is recommended by the Association of Public Health (APHA).
Potato-Extract-Glucose Agar can be used in the analysis of dairy products, bottled drinks, frozen food,
and other types of food. It can also be used in the identification of fungi and yeasts in parallel with their
cellular morphology or in methods of microcultivation in slides.
When the medium is to be used for enumeration of molds and yeasts, add to the medium, sterilized and
cooled to 45-50 C, approximately 14 ml of a sterilized 10 % solution of tartaric acid to obtain a pH of 3.5.
Do not heat the medium after adding the acid, because the agar may hydrolyze and not solidify.
MICROBIOLOGICAL TEST
The following results were obtained in the performance of the given medium from type cultures after
incubation at 35±2 °C and observed after 18-24 hours.
