HERSTELLUNG
62 g des Mediums werden in einem Liter destillierten Wassers suspendiert. Gut mischen. Man erhitze bis zum Kochen,
um das Medium vollständig zu lösen. Man gebe die Lösung in geeignete Behälter und sterilisiere 12 Minuten lang bei
121°C.
EINSATZGEBIET
MRS Agar wurde von deMan, Rogosa und Sharpe entwickelt, um alle Stämme von Lactobacilli zu kultivieren,
insbesondere die anspruchsvollen, langsamer wachsenden Typen wie L. brevis und L. fermenti. Das Medium ist geeignet
zur Kultur von Milchsäurebakterien im allgemeinen, einschließlich Lactobacillus, Streptococcus, Pediococcus und
Leuconostoc. Ammoniumcitrat inhibiert bei niedrigem pH die meisten Mikroorganismen, einschließlich anderer
Streptokokken und Schimmelpilze. Es erlaubt das Wachstum von Lactobacillus, aber es unterbindet das Ausschwärmen im
Allgemeinen.
Lactobacilli sind mikroaerophyl und erfordern normalerweise ein Ausstreichen auf zwei Schichten. Die
Oberflächenkolonien oder die eingebetteten Kolonien sind normalerweise klein, trüb und weiß, dicht und federig. Die
Platten werden gegossen, indem zuerst 1 ml des vorher gelösten Musters auf eine sterile Petrischale gegossen wird und
dann das abgekühlte Medium (45-50 °C) zugefügt wird. Nach Festwerden wird eine zweite Schicht gegossen. Die
Platten werden 3 Tage lang bei 35 °C, oder besser 5 Tage lang bei 30 °C inkubiert. Es ist wichtig, dass für eine feuchte
Atmosphäre gesorgt wird, da die Platten während der Inkubation unter 5 % CO2 nicht austrocknen sollten.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Medium / Agar bei
einer Temperatur von 35 ± 2 °C für 48 h und 5 Tage.

PREPARATION
Suspend 62 g of the medium in one liter of deionized or distilled water. Mix well. In order to dissolve the medium completely
heat to boiling. Dispense and sterilize at 121 C for 12 minutes.
USES
MRS Agar was developed by deMan, Rogosa and Sharpe to grow luxuriantly all strains of lactobacilli especially the
fastidious, slower-growing types such as L. brevis and L. fermenti. The medium is apt for the growth of lactic acid bacteria,
including Lactobacillus, Streptococcus, Pediococcus and Leuconostoc. Ammonium citrate at a low pH inhibits most
microorganisms, including other Streptococci and molds. It allows for the growth of Lactobacillus, but limits swarming in
general.
Lactobacilli are microaerophilic and generally require plating of two layers. The surface colonies or the embedded colonies
usually are small, opaque and white, dense and feathery. The pour plate method deposits 1 ml of the previously diluted
sample into a sterile Petri dish and the cooled (45-50 C) medium is added. After solidification, a second layer is poured.
The plates are incubated at 35 C for 3 days or better, at 30 C for 5 days. It is important to maintain a humid atmosphere
because the plates should not dry out during incubation which is in 5 % CO2.
MICROBIOLOGICAL TEST
The following results were obtained in the performance of the given medium from type cultures after incubation at a
temperature of 35 ± 2 °C and observed after 48 hours and after 5 days.
