CASO Agar ist ein sehr nährstoffreiches Medium für den allgemeinen Einsatz im mikrobiologischen Labor. Es unterstützt das üppige Wachstum von anspruchsvollen Organismen wie Pneumococcus, Streptococcus, Neisseria etc. Es ist äußerst hilfreich bei der Bestimmung von hämolytischen Reaktionen.

HERSTELLUNG
40,0 g des Mediums werden in einem Liter destillierten Wasser suspendiert. Man erhitze unter häufigem Rühren/Schütteln und lasse eine Minute lang kochen. Das Medium muss vollständig gelöst sein. Man sterilisiere im Autoklaven 15 min (nach Ph. Eur.) bis 20 min bei einer Temperatur zwischen 118 und 121 °C. Wenn größere Mengen angesetzt werden, muss die Sterilisationszeit, jedoch nicht die Temperatur oder der Druck erhöht werden. Man lasse abkühlen und gieße in Petrischalen. Für hämolytische Untersuchungen gebe man 5 bis 10 % Schafs- oder Kaninchenblut zu.
EINSATZGEBIET
CASO-Agar wird empfohlen von der Pharmacopeia Europaea zur Prüfung der untersuchten Produkte auf Anwesenheit von Bakterien mittels Membranfilter-Methode, sowie zur Zählung der Bakterien auf Agarplatten oder mit Hilfe der Verdünnungsmethode (MPN-Methode). Da dieses Medium zwei durch enzymatische Hydrolyse von Casein- und Sojaprotein erhaltene Peptone enthält, unterstützt es eine große Palette an Mikroorganismen, einschließlich anspruchsvoller aerober und anaerober Organismen. Da es keine Kohlenhydrate enthält, ist es sehr nützlich bei der Untersuchung von hämolytischen Reaktionen und zur Herstellung von Schokoladenagar. Wenn dies gewünscht wird, können leicht Antibiotika sowie andere Zusätze oder Inhibitoren zugesetzt werden. Zu den Mikroorganismen, die unter vielen anderen in diesem Medium wachsen, gehören: Streptococcus, Neisseria, Brucella, Corynebacterium, Listeria, Pasteurella, Vibrio, Haemophilus vaginalis, Candida, etc.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Medium / Agar bei einer Temperatur von 30-35 °C für 18-24 Stunden. (Agarplatten mit oder ohne Blut)

CASO Agar is a medium very rich in nutrients for general use in microbiological laboratories. It supports the abundant growth of fastidious organisms such as Pneumococcus, Streptococcus, Neisseria, etc. It is very useful for determination of hemolytic reactions.

PREPARATION
Suspend 40 g of the medium in one liter of deionised or distilled water. Heat with frequent agitation and boil for one minute until complete dissolution. Sterilize in an autoclave between 118 and 121 C for 15 (acc. to Ph. Eur.) to 20 minutes. In the case of large volume preparation, increase the time of sterilisation but not the temperature or pressure. Cool and pour into Petri dishes. For hemolytic studies, add 5 to 10% sheep or rabbit blood.
USES
CASO-Agar is recommended by the Pharmacopeia Europaea for examination of samples by the membrane-filter method for the presence of bacteria. Furthermore, it is recommended for counting bacteria on agar plates or by dilution series. Containing two peptones obtained by enzymatic hydrolysis of casein and soy protein, this medium supports the growth of a great variety of microorganisms, including fastidious aerobes and anaerobes. Since it lacks carbohydrates it is very useful in the study of hemolytic reactions and also in the preparation of chocolate agar. If desired, antibiotics can easily be incorporated as well as other supplements or inhibitory agents. Typical microorgansisms growing on this medium are: Streptococcus, Neisseria, Brucella, Corynebacterium, Listeria, Pasteurella, Vibrio, Haemophilus vaginalis, Candida, etc.
