HERSTELLUNG (EN ISO 6222:1999 – 7.3.)
24 g des Mediums werden in 1 l destilliertem Wasser suspendiert. Unter Rühren zum Kochen erhitzen. Das
Medium muss vollständig gelöst sein. Für 15 Minuten im Autoklaven bei 121 °C sterilisieren. In Aliquots zu
15 oder 20 ml abfüllen und erstarren lassen. Die Aliquots müssen bei 4-6 °C aufbewahrt und innerhalb von
1 Monat verbraucht werden. Das Medium vor Gebrauch schmelzen, abkühlen lassen und in einem
Wasserbad bei ca. 45 °C für maximal 4 Stunden aufbewahren.
EINSATZGEBIET
Hefeextrakt-Agar wird vorgegeben von der Europäischen ISO Norm 6222:1999 zur Untersuchung von
Wasserproben auf die Anwesenheit von kultivierbaren Mikroorganismen nach aerober Inkubation bei 36 °C
und 22 °C und zur Zählung dieser Mikroorganismen. Petrischalen werden mittels der Platten-GießMethode angeimpft (ISO 8199). Hierzu gibt man in eine trockene Petrischale bis zu 2 ml der Probe und
gießt nach maximal 15 Minuten 15 bis 20 ml des geschmolzenen und abgekühlten Mediums hinzu. Durch
leichtes Schwenken mischen und das Medium stocken lassen. Pro Probe mindestens zwei Platten
animpfen und je eine Platte umgedreht bei 22 °C für etwa 68 Stunden und eine Platte umgedreht bei 36 °C
für etwa 44 Stunden inkubieren. Es empfiehlt sich ebenfalls, einige Probenverdünnungen anzuimpfen, um
in jedem Fall einen zählbare Platte zu erhalten. Kulturfähige Mikroorganismen im Sinne der Norm sind alle
aeroben Bakterien, Hefen oder Schimmelpilze, die in Hefeextrakt-Agar unter den vorgegebenen
Bedingungen Kolonien formen können.
Die Europäische ISO Norm 6222:1999 regelt die Überprüfung der Effizienz der Entkeimung von
öffentlichem Trinkwasser und generell der Keimzahlen in allen Wässern. Die Methode ist grundsätzlich
geeignet zur Untersuchungen von jedem Wasser für den menschlichen Gebrauch. Bitte ziehen Sie hierzu
auch die Vorgaben der folgenden Normen hinzu: EN ISO 3696, EN ISO 5667-3, EN 255667-2, ISO 6887,
ISO 8199.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Agar
bei einer Temperatur von 36 ± 2 °C für 44 ± 4 Stunden und bei einer Temperatur von 22 ± 2 °C für
68 ± 4 Stunden.

YEAST EXTRACT AGAR

PREPARATION (EN ISO 6222:1999 – 7.3.)
Suspend 24 g of the medium in 1 l distilled water. Heat with frequent agitation until complete dissolution.
Sterilise for 15 minutes at 121 °C in an autoclave. Distribute volumes of 15 to 20 ml in tubes or bottles and
let cool. The aliquots have to be stored in the dark at 4-6 °C. Do not use longer than 1 month. For use, melt
the medium, allow to cool, and maintain it at approx. 45 °C in a water bath for no longer than 4 hours.
USES
Yeast Extract Agar is recommended by the European ISO Norm 6222:1999 for the examination of water
probes for cultivatable micro-organisms by aerobic Incubation at 36 °C and 22 °C, as well as for the
enumeration of these micro-organisms. Petri dishes are inoculated by the pour-plate method (acc. ISO
8199). Place a volume of the test sample not exceeding 2 ml in the Petri dish, add 15 to 20 ml of the molten
and cooled medium and mix carefully by gentle rotation. Allow the medium to set. Time between addition of
the test sample and addition of the medium shall not exceed 15 min. Inoculate at least 2 plates per sample
and incubate 1 set at 22 °C for approx. 68 hours, the other set of plates at 36 °C for approx. 44 hours. We
also recommend inoculation of all samples in several dilutions, in order to gain at least one set of plates that
can be counted. Cultivatable microorganisms as mentioned by the ISO norm are all aerobic bacteria, yeasts
and moulds capable of forming colonies in Yeast Extract Agar under the test conditions described therein.
The European ISO Norm 6222:1999 is intended to measure the operational efficiency of the treatment
process of public drinking water supplies and for general application of all types of water. It is particularly
applicable to the examination of water intended for human consumption. See also: EN ISO 3696,

MICROBIOLOGICAL TEST
The following results were obtained in the performance of the given medium from type cultures after
incubation at a temperature of 36 ± 2 °C and observed after 44 ± 4 hours and at a temperature of 22 ± 2 °C
and observed after 68 ± 4 hours.
