BLUTAGAR (BASIS)
Zur Isolierung, Kultivierung und zum Nachweis von hämolytischen Reaktionen anspruchsvoller Mikroorganismen
Blutagar (Basis), eignet sich für die Isolierung und Kultivierung einer Reihe von Mikroorganismen, die andernfalls schwierig anzuziehen sind. Nach Zugabe von Blut kann er zur Bestimmung hämolytischer Reaktionen eingesetzt werden.

HERSTELLUNG
40 g des Mediums werden in einem Liter destillierten Wassers suspendiert. Nach einer Ruhepause von 5 Min. rühre man, bis eine gleichmäßige Suspension entstanden ist. Man erhitze unter vorsichtigem Rühren/Schütteln und lasse eine Minute lang kochen. Man sterilisiere 15 Min. lang im Autoklaven bei 121 °C. Man lasse auf 45 – 50 °C abkühlen, gebe 5 bis 10 % steriles defibriniertes Schafs- oder Kaninchenblut hinzu, mische vorsichtig und gieße in sterile Platten. Wir empfehlen die Verwendung von Schafs- oder Kaninchenblut.
EINSATZGEBIET
Sobald das Medium geschmolzen und auf 45 °C abgekühlt wurde, können 5 – 10 % defibrinierten sterilen Schafsbluts zugegeben werden, womit der Haemophilus wettgemacht werden kann. Die Bildung von Blasen sollte möglichst vermieden werden, wenn das Blut zu dem abgekühlten Medium zugegeben wird, danach rotiere man den Kolben oder die Flasche langsam, um eine homogene Lösung herzustellen. Das Medium kann dann in Petrischalen gegossen werden und erstarren. Die leere Petrischale kann auch mit einer geringen Menge des Probematerials inokuliert werden und dann das Medium bei 50 °C zugegeben werden; die Platte sollte dann vorsichtig geschüttelt werden, um das Inokulum zu homogenisieren. In einigen Labors wird das Medium in Röhrchen mit Schraubkappen zubereitet, die bei 45 °C inokuliert und dann in sterile Petrischalen gegossen werden können.
MIKROBIOLOGISCHE TESTS
Die folgenden Ergebnisse wurden erzielt nach Inkubation von Referenzstämmen im angegebenen Medium / Agar mit 5 % defibriniertem Schafsblut bei einer Temperatur von 35 ± 2 °C für 18 – 24 Stunden.

For the isolation, cultivation and detection of hemolytic reaction of fastidious microorganisms
Blood Agar (Base) is suitable to isolate and cultivate a wide range of microorganisms with difficult growth. Upon adding blood, it can be utilized for determining hemolytic reactions.
Approximate formula in g/l:
Heart Infusion ............................ 10.0

Meat Peptone ............................ 10.0

PREPARATION
Suspend 40 g of the medium in one liter of deionized or distilled water. Allow to stand for 5 minutes and mix well until a uniform suspension is obtained. Heat with gentle agitation and boil for one minute. Sterilize in an autoclave at 121 °C for 15 minutes. Cool to 45-50 °C, add 5 to 10 % of sterile defibrinated sheep or rabbit blood, mix carefully and pour into sterile plates. Human blood is not recommended. Use preferably sheep or rabbit blood.
USES
Once the medium has been melted and cooled to 45 ºC you can add 5-10 % of defibrinated sterile sheep blood, in this case you can recuperate Haemophilus. Be careful to avoid bubble formation when adding the blood to the cooled medium and rotate the flask or bottle slowly to create a homogeneous solution. The medium can then be poured into dishes and solidified.
You can also inoculate the empty Petri dish with a small amount of specimen material and then pour the medium at 50 °C, swirl the plate gently to homogenize the inoculum. In some laboratories the medium is prepared in screw-capped tubes which can be inoculated at 45 °C and then poured into sterile Petri dishes.
MICROBIOLOGICAL TEST
The following results were obtained in the performance of the given medium from type cultures, with 5 % of defibrinated sheep blood, after incubation at a temperature of 35 ± 2 °C and observed after 18 – 24 hours.
