Modifizierte Färbung nach Pappenheim

Mit dieser Färbemethode werden üblicherweise Blut- und Knochenmarksausstriche dargestellt.
Sie kann aber auch allgemein als schnelle Übersichtsfärbung verwendet werden.

Der eigentliche Färbevorgang ist nach ca. 1 Minute abgeschlossen.

Kit enthält

 Färbelösung 1 (Best.-Nr. 8934) 250 ml / 500 ml
 Färbelösung 2 (Best.-Nr. 8935) 250 ml / 500 ml
 Fixativ (Best.-Nr. 8939) 250 ml / 500 ml

Lösungen vor Gebrauch filtrieren! Die Lösungen können nicht einzeln nachbestellt werden.

Material:

 Hitzefixierte, luftgetrocknete zytologische Präparate wie Blut- und Knochenmarksausstriche,

aber auch Urin, Sputum etc.

 Formalinfixierte, entparaffinierte Gewebeschnitte

Durchführung:

1. Inkubieren in Fixativ 5 sec

5. Färben in Lösung 2 10 sec

2. Lösung abtropfen lassen

6. Lösung abtropfen lassen

3. Färben in Lösung 1 10 sec

7. Spülen in Leitungswasser 10 sec

4. Lösung abtropfen lassen

8. Entwässern (optional) und eindecken

Ergebnis:

 Erythrozyten: rosa bis gelbrot
 Thrombozyten: violett bis purpur
 Monozyten:

violette Kerne, hellblaues Zytoplasma

Man beachte:

Die Farbintensität ist abhängig von der Vorbehandlung und Beschaffenheit der zu färbenden Probe.
Es kann also ggf. nötig sein, die Methode an die jeweiligen Bedingungen anzupassen.

Panoptical Fast Staining Kit

Modified Staining acc. to Pappenheim

The staining method is typically used for visualising the different elements of blood and bone marrow smears.
But it can also be applied as fast oversight staining.

The staining process is completed after ca. 1 minute.

Kit includes:

 Staining solution 1 (Art.-No. 8934) 250 ml / 500 ml
 Staining solution 2 (Art.-No. 8935) 250 ml / 500 ml
 Fixatif (Art.-No. 8939) 250 ml / 500 ml

The staining solutions should be filtrated before use! Solutions cannot be bought separately.

Material:

 Heat-fixed, air-dried cytological specimens like blood and bone marrow smears,

but also urine, sputum etc.

 Formalin fixed, de-waxed tissue sections

Instruction:

1. Incubate in fixative 5 sec

5. Stain in solution 2 10 sec

2. Let the solution drip off

6. Let the solution drip off

3. Stain in solution 1 10 sec

7. Rinse in tap water. 10 sec

4. Let the solution drip off

8. Dehydrate (optional) and mount the specimen

Result:

 Erythrozytes: rose to yellow-red
 Thrombozytes: violet to purple
 Monozytes:

violet nuclei, light blue cytoplasm

Please note: The colour intensity depends on the pre-treatment and the composition of the samples to be
stained. It may initially be necessary to adapt the method to the respective conditions.
