The Tissue-Tek® GlasTM Coverslipper is a system that dispenses mounting medium onto stained histopathological and cytopathological samples on glass slides as well as coverslipping these samples using cover glasses.

Preparation before cover slipping
Hardware
Placing cover glasses
1. Put cover glass holder in place.
2. Make sure the tab of the cover glass holder fits
in the holder (check on the CG size).

When setting the mounting medium bottle for the first time, you must fill the discharging channel with mounting medium.
1. Place the waste bottle below the dispensing
nozzle.
2. Fill the priming bottle with mounting medium.
3. Connect the tip of the introduction tube with
the priming bottle.
4. Squeeze the priming bottle to feed the
mounting media until it reaches the dispensing nozzle.
5. Remove the priming bottle from the
introduction tube.
6. Set the mounting medium bottle.
7. Tilt the mounting medium tube towards the
front and lift the tube.
8. In order to prevent air bubbles, insert the tube
gently into the bottle.
9. Perform an [initial priming] page 2.7 OM.
10. Press [EMER. STOP] to enter the set up
menu.
11. Highlight [1. Prime Dispenser].
12. Press [Verify].
13. Press [Start] and again [Start] to prime.
Changing mounting medium brands Since mounting mediums vary in viscosity and formulation, it will be necessary to follow the same priming procedure when changing to a different brand of mounting medium.
Follow the detailed instructions written in the Operating Manual, “priming the instrument” beginning on page 2.7.

Installing the nozzle reservoir
1. Set the dispensing tray in the system by
aligning the tray with the two guide rails (only for the first time use).

1. Fill the anti drying bottle with solvent to just
below the lid.
Note: type of solvent depends on the mounting medium
2. Place the anti-drying bottle and waste
container in the dispensing tray.
Handling the dispensing nozzle
1. When setting the anti-drying bottle, push down
the dispensing nozzle in a backward direction and then set the bottle.
2. After the anti-drying bottle and waste container
have been set, lift the dispensing nozzle and return the nozzle so that its tip enters the hole in the anti-drying bottle. (see description “handling the dispensing nozzle”).

1. Remove the lid and add solvent for mounting
medium into the pump reservoir.

2. Fill the pump anti drying bottle with solvent to

Preparation before coverslipping
Software VOLUME KNOB
Can be used to fine tune the volume of mounting medium dispensed on the slide. SPEED KNOB Refers to the speed at which the coverglass is applied to the slide.
STOP
Can be used to stop the instrument while operating.
TEST
Can be beneficial if the operator would like to test one slide.
SIZE
Toggles between coverglass sizes 40, 50, 55 and 60 mm.
VERIFY
Key acts as an enter key.
PRIMING FUNCTION
Can be used to verify proper dispensing. When single key is pressed mounting medium will be dispensed from the nozzle one time. When continuous key is pressed the mounting medium will dispensed continuously until the “STOP” key is pressed.

Pre-operational checks
1. Turn on POWER.
2. Remove the loading station lid.
3. Check the solvent volume level in anti-drying
bottle and dispensing pump reservoir.
4. Clean any possibly spilled mounting medium.
5. Place receiving racks in the unloading area.
6. Put coverglass (CG) holders in place and fill
with coverglasses.
7. Check if CG size on screen matches CG in
holder.
8. Check the volume level of mounting medium.
Start of operation
1. Check CG size, coverslipping speed and volume
of mounting medium.
2. Select coverslipping program.
3. Press and hold [prime] key for 3 seconds
4. Pull out the loading station and set baskets.
Close loading station door and [start] coverslipping.

Maintenance
(section 5 operating manual) Daily
Station cleaning
1. Clean each station and its surrounding area.
2. Brush off each station, remove dust and/or
glass particles.
3. Using a cloth moistened with solvent for
mounting medium*, remove any mounting medium attached to the stations.
Cover glass and Slide glass arm cleaning
1. Clean the suction pads using a cloth moistened
with 100% alcohol.
End of Operation
1. Remove receiving racks.
2. Remove empty baskets.
3. Place dispensing nozzle in the lubrication bottle
above the solvent (1)! If the instrument will not be used for more than one week, place the dispensing nozzle in the solvent (2) by pulling the lever towards you.
Power off
1. Turn off power and place the lid on the loading
station.
Mounting medium nozzle cleaning
1. Clean the mounting medium nozzle by using a
cloth moistened with solvent for mounting medium*.
2. At the end of every day before switching off the
machine, place dispensing nozzle in the anti­drying bottle above the solvent!
Weekly Waste container
1. Remove waste container and replace it with a
clean container.
2. Check the spatula part. Soak the used waste container for 24 hours in solvent for mounting medium*. Cleaning the loading station

6 Months Anti-drying bottle
1. Change the solvent for mounting medium* in
the anti-drying bottle.
Cleaning the receiving racks
1. Soak the racks for 24 hours in solvent for
mounting medium*. Instrument strainer Open cover on the right side of the instrument and remove the strainer cap. Use xylene soaked brush to clean the filter. Replace the cap.
Yearly
1. Preventive maintenance by a Sakura
representative or a qualified service engineer.

The trouble shooting part of the operating manual is divided into three sections to explain how to recover from conditions that may occur during routine operation.
In the first section,
Troubleshooting Chart is provided, which identifies possible conditions related to the electrical and mechanical operations.

Messages, explains specific messages that may be displayed during routine operation in detail.
The third section,
Gives a detailed explanation of all error codes and how to recover from them.
Operating manual section 6, trouble shooting
1. Change the solvent for mounting medium* in
the loading station.
* Solvent for mounting medium is the reagent which is compatible
with the mounting medium, Tissue clear when using Tissue Mount
and Xylene when using Glass coverslipping medium.
