Streptococcus mutans is a Gram-positive faculatively anaerobic bacterium with a genome
of over 2Mbp. The genome is arranged in a circular conformation with 3 separate plasmids
of approximately 5Kbp each. This bacterium thrives between 18 and 40°C and can be
found inhabiting the oral cavity of almost all humans.
S. mutans in the oral cavity bind to the enamel of teeth and replicate to form a biofilm. The
layer of bacteria metabolise carbohydrates into glucose and fructose with the former
allowing the bacteria to attach to the enamel surface via adhesive glucans and the latter
providing energy for bacterial replication. The fermentation of these sugars produces an
acidic environment in the mouth that decalcifies the tooth enamel resulting in cavities and
potentially tooth loss. The formation of the biofilm makes this bacterium difficult to treat
with antibiotics as their antibiotic tolerance can increase dramatically.
Introduction to Streptococcus mutans
The glucosyltransferase-I gene has previously been identified as a highly specific market
for S. mutans (Lett Appl Microbiol. 2006 Feb; 42(2):127-31). The primers and probe have
100% homology with all reference sequences for S. mutans in the NCBI database.
