STEVE
STEVE i s Nanolive’s softwa re for exploring the da ta acquired using th e 3D Cell Explorer. For
each recorde d frame, the 3D Cell Exp lorer measures the Ref ractive Index of yo ur cell in three
dimensions . Use STEVE to digitally stain your data and interactively explore your cell and its
subcomponents
2D VIEW
The 2D view show s an X-Y slice of the sample’s Ref ractive Index over laid with the user- defined
stains. I n this view you can: Move t o a different slice ( or via Sl ices slider) • Zoom in or out (
Move the view w hen zoomed in (SHIF T + ).
: Measure (or stop meas uring) a user-defined p ath length. Click t his button and click o n
the image to dr aw a path • : Show the Refrac tive Index and the I ndex Gradient in the Pa nel
Viewer of a sele cted voxel in the 2DView • : Take a screenshot of the 2D view • / : Switch
between St andard mode and Vi ewer mode.
PANEL VIEWER
The panel view er represents a 2D space of Ref ractive Index (ho rizontal axis) and Index Gradient
(vertical a xis). Stains are shown as r ectangles coverin g a region in the staini ng space. Regions
on the right ha ve a high Refractive I ndex, regions on the t op have a high Index Gradi ent.
In this view yo u can: Change a stain’s pos ition ( on the stain’s cente r) or shape ( on the
stain’s edge) • Chan ge a stain’s transparenc y ( on the stain’s center/edge o r via the Opacity
and Edge Sof tness sliders).
3D VIEW
The 3D view show s the stained data in t hree dimensions. In t his view you can rotat e ( ) •
Zoom ( ) • Move (SHIFT + ) the 3D dat a.

BUTTONS AREA
: Visuali ze your sample in white li ght. Move the micros cope’s knobs to search an d focus your
cell of interest • : S ingle shot acquisit ion: acquire a single 3D fr ame • : Time-lapse
acquisitio n: acquire multiple 3D fra mes in a range of time • : Load a measurement file (.vol) •
: Save a 3D or 4D mea surement file (.vol). To save a 4D file, pleas e indicate the desi red
range on the t ime line using marker s and • : Open the Ex port dialog.
SLIDER S AREA
Overlay : Fuzzy switch bet ween stained dat a and unaltered Refrac tive Index.
Slices: Se lects the X-Y slice from t he 3D data stack.
Opacity : Chooses the opa city for the curre ntly selected stain .
Edge Softn ess: Use this to make a st ain’s edges soft and avoid s olarized images in 2D.
STAIN COLORI NG AREA (PANE L)
Load Panel: L oad a previously save d panel (.xml format )
Save Panel: S ave the current pan el (stains)
Rescale: Mo dify the viewin g range of the panel vie wer to fit the current ti me frame.
STAIN PICKER
: New stain - choose a n ame (not required) and a c olor for the stain. Ne xt, begin painting
your region of i nterest in the 2D view ( ). A rectangular st ain is computed based o n the
painted par t of the image and is show n in the panel viewer.
/ : Hide or show a st ain • : Delete a stain • : Show t he stain properti es and allow
background definition.
3D CONTROL A REA
: Change the ba ckground color for the 3D v iew.
Hide Axes & Cu be: Show/hide the coo rdinate axes and the ou tline cube in the 3D vie w.
Show Capti on: Show/hide the l abels in the 3D view.
Cropping sph ere: Cut away part of the 3D v iew. Move the plane thr ough the sphere to crop
the 3D data. T he plane can be move d (SHIFT + ) or rotat ed ( ).
TRANSPORT CONTROL
Use this to exp lore your data in time. T he transport displ ay shows the frame n umber and
acquisitio n time of the current f rame.
: Jump to t he beginning of a data s et • : Set marker A to th e current cursor po sition •
: Play/paus e playback • : S et marker B to the current c ursor position • : Jump to the en d
of a data set • : Set playb ack speed. (Note: Ma rkers A and B select a tim e range for saving /
exporting purposes.)
TIME LINE AREA
Displays t he current time fram e ( ) and the select ed range ( ).
Click on the t op part of the area to jump t o the specified fram e. Drag on the tick regi on to set
markers A and B .
