Introduction
Thank you for purch asing UV/VIS S pectrophotometer, Optizen Series.
Optizen Serie s can be applied var ious elds from a simple examinatio n like a water quality
analysis to a comp lex examination like a bio -chemic al analysis. User c an measure easier,
faster, and more accurate by c onvenient interf ace and auto- function.
In addition, user c an check and c ontrol experimental results on th e real time by OptizenView,
PC-Inter face sof tware of Optizen S eries.
Using Manual
This manual includes the ways of system installation, Optizen operating.
The manual is com posed of four par ts, which are c ategorized upo n the subjects; the refore,
user can searc h quickly the nec essary par ts.

1. Turning on Switch
It is self-tunin g about CPU & ROM, Wave Moter, Cell Moter, Filter Moter, W Lamp, D2 Lamp
and D2 Wave. Results of self-t uning indicate Ok o r Error. Incase Error o ccurs at any step,
self-tuning is s topped. If you want to che ck next step, push [ENT ER] button. Only ever ything
is OK, automatically move to Main Mo de.
To get more stable data, pleas e warm up the system abou t 30 minutes afte r turning on the
switch.

You can change setup by selecting number.
1) Cell Mode
You can setup about Cell Mo de. Default is Multi c ell Mode. If you want to use S ingle cell
holder, have to change cell m ode to Single cell holder.
Multi Cell Mode : Use 8 hole cell ho lder.
Single Cell Mode : Use single Cell H older.
2) Unit
To change unit, push number 2 th en show up 12 concentrat ion unit. After moving with Up/
Down button, c onrm with [Ente r] button then uni t is changed. Default is CONC.
3. MAIN MENU (Selecting Mode)
Real time cloc k function is supp orted at only 2120UV p lus
Move to measuring mo de by selecting number key.
To move another mode after measuring one mode, c omeback to this ‘ Main Menu’ then se lect
num ber.
Even though mode is ch anging, measured data remain without saving until turn of f Optizen.
On the upper lef t side is a title an d on the upper right s ide shows data infor mation, print
status, and the c ell position st atus. The central s creen shows the gr aphs or detail infor mation
on the measured dat a. The bottom si de of screen indic ates explanation, c ommand, and the
choice box for f unction.
4. Cell Moving Method
Cell Holder is d esigned to hold up to 8 C ells. Each Cell h as number “B, 1, 2, 3, 4, 5, 6, 7 ” and
it is possible to m ove a cell upward and downwa rd respectively by [ UP], [DOWN].
Please keep in mind that t he rst cell, “ B” is assigned to p ut Reference Cell.

9) Calibration Mode (Secret Key:’9’)
Optizen has self- correc tion mode for wavele ngth and cell hol der position. Wavelen gth
calibration mode checks the 0 th order posit ion of grating and th e peak wavelength (656 .1nm)
of Deuterium lamp.

Wavelength Tuning : Self-correction f or wavelength.

Warning : It is very strongly re commended th at ONLY expert or fully trai ned staff
engineers ope rate these advanced applicatio ns. Fatal errors c an be caused by
some incorrect operations.
3) D2 Lamp Save Mode
It automatical ly turns off the p ower of the D2 lamp that is n ot used by selecting number 3.
Incase this mo de ‘ON’, D2 Lamp is off at our o f wavelength range.
‘Wait a moment ’ message is showe d up at that time. A few seconds is need.
The wavelength rang e of Optizen 2120UV spect rophotometer is f rom ultraviolet ra diation
(190nm) to near infrared radiati on (1100nm). For this, deuteriu m lamp and tungsten -
halogen lamp is us ed as a light source. Two lamps are ch anged each other au tomatically
in 350~ 470nm range.
4) LCD Contrast Control
It controls br ightness level of LCD bac k light.
5) Lamp Change WL.
You can setup the wavelength for c hanging point of deuterium lamp and tungsten-halog en
lamp.
The range of the valu e that you can input is f rom 350nm to 470nm.
The default value is 370nm.
6) Initial Wavelength
You can set initial wavelength of Optizen. Incase set mainly used wavelength, you don’t need
to set wavelength every measuring time.
7) Clear Lamp Position
Clear Lamp posi tion of W Lamp and D2 Lamp.
8) Set Date & Time (Only 2120UV Plus)

You can setup wavelength again or m easure AUTOZERO [F3] again.
With Standard Curve
1. At Mode setup step, you can sel ect and open a saved St andard Curve.
Ex) Incase you want to use Standard Curve AAA A in ST0, input No. 0.
2. Enter the CELL [F3] th en select the ce ll to measure.
3. MEASUR E [F2], Measured re sult is like the top right side gure.
4. Can measure again w ith MEASUR E [F2] and delete all dat a with DELETE [F4].

2. Survey Scan
1) Initial Screen of Test Mode
At this mode, you ca n survey scan abou t a sample in selected wavelength range. You can
see Absorba nce or Transmittance d ata with Graph or Table.
At the Survey Sc an Mode screen, you w ill nd 'New bas eline is needed' c ommand.
BASELINE is like a Bla nk of ATC Mode. You must measure BASELINE [F1] rst.
<Main Screen>
Baseline [F1] : On the proce ss of Baseline Sca nning, its status is i ndicated by '%'. When
the Baseline Sc an is over, the 'READY' sign will come up.
New Scan [F2]
Select the ce ll you intend to measure an d
push NEW SCAN [ F2] button, then it w ill scan
the wavelength sele cted in each cel l.
While survey sc an, you can see Abs orbance
or %Transmittance as wavelength on t he
upper side of scre en. The middle par t of
screen will indi cate real time gra phs.
If the scanning i s over, the graphs, which are
automatically readjusted to the out put range
of Y, will be drawn promptly. Based upon th is
data, all the gur es for graphs and tex ts of
Survey Scan M ode will be determi ned.

ABS / %T [F4]
It is a Toggle Switch which d ecides whethe r to view the data in Ab sorbance or in
%Transmittance. The range of wavelength is maintained during toggling.
2) Mode Setup
Start Wavelength : Input start waveleng th to survey scan. Default value is 400.0 n m.
Finish Wavelength : Input nish wavelength to sur vey scan. Default val ue is 700.0 nm.
Input wavelength at ‘S et new wavelength _ nm’ with num ber key.
3) Measure step by step
1. Setup wavelength to measure at M ode setup step.
2. Select the c ell to measure with U p/Down butto n.
3. Measure Base Lin e [F1].
4. Measure New Scan [F2].
5. Conrm dat a with Graph, Ab s/%T.

select the ran ge of wavelength. The modied
graphs by this mode w ill be kept.

At ABS/%T/CONC mode, you c an get concentr ation of samples us ing a saved standard
curve. Open st andard cur ve name is indicated at u pper side of scree n.

Wavelength : Setup wavelength to measure. In case select num ber 1, the message of ‘Set
new wavelength _ nm’ shows up. H ere input wavelength wi th number key
then [ENTER].
Select Type : Select the type of Sta ndard Curve you pla n to make. There are three k inds of
Curve t ype.
- Linear(0) Standard C urve : Linear ty pe that passes zero point
- Linear(N) Standard Curve : Gener al linear type
- Segmented Stan dard Curve : A Type of graphs that l inks certai n points.

1) Initial Screen of Test Mode
At initial scree n, you can see only AD D [F1], FACTOR [F4]. To make a standard curve, select
ADD [F1] and input concent ration. Then ME ASURE [F2] wi ll appear. With FACTOR [F4] you
can add data at saved st andard curve. You can also FACTOR OPEN or DELETE.
At this mode you ca n make a standard cur ve with standar d solution. You can save standar d
curve by ten.

To make standard curve, you have to measure minimum 2~3 standa rd solutions.
MEASURE [F2] : Measure conc entration of sta ndard solution. A fter measuri ng, you can
add another con centration by AD D [F1] and measure again by MEASU RE
[F2].

Single cell Mo de : After measuring Reference t hen insert st andard solution a nd measure
in order.
After measur ing, can add concentration wi th ADD [F1].
4. You can conrm measured data with GR APH [F3].

- NORMAL :
Incase make a stand ard curve by measur ing concent ration of standar d solutions.
- INPUT : By inputting C oncentration a nd Absorbanc e by maximum 30, you c an make
Standard Cur ve. After input ting, you can make sta ndard curve auto matically by
clicking ME ASURE [F2] in Test Mode.
Data Clear : Incase selec t the [Data Clear], you can see the mes sage ‘Warning! ! All data
will be lost. Pres s [ENTER] to Conrm.’ then all data is deleted.
3) Measure with Multi cell
1. Setup wavelength and graph t ype at Mode Setu p step.
2. Select AD D [F1] at initial screen of standar d mode then input c oncentration o f standard
solution in order.
You can add by 7 concentrati ons. After addi ng over 2, then shows up ME ASURE [F2] key.
Incase select Input mode
Select
ADD [F1] then ‘Set concentration: _’ shows up. Here input concentrat ion then [ENTER].
After that, t he message of ‘Set a bsorbance: _’ shows up t hen input absorb ance. Conrm
with [ENTER]. Repeat as number of sample s.

User input Factor and Wavelength at mode setup step t hen ABS Ratio is me asured and
calculated auto matically. The ratio means A
260nm/A280nm
.
1) Test Mode initial screen
It automatical ly calculates the ratio per 2 or 3 Wavelength s.
Start the m easurement by click ing MEASUR E [F2] in the beginning. Prior to measurement,
please input the Wavelength, Cell Number a nd Factor at the Mode Set up.
You can measure maximum 7 sa mples.
More time is nee ded compare wit h other mode, bec ause measure at 2~3 wavelengt h
about one sample.

TN-C : TN means total measuri ng times, C means Ce ll number.
ABS1 : Measured absorbanc e at Wavelength 1 (260nm).
ABS2 : Measured absorbanc e at Wavelength 2 (280nm).
RATIO : The ratio ABS 1and A BS 2
RESULT : Ratio result considere d factor Ratio. Inc ase no factor; it ’s a same value with ratio
column.

MEASURE [F2]

Number and Factor at the Mode Setup.
The result is indic ated like the left side gure.

Reference Wavelength : Wavelength that will measure Reference, and default i s 320nm.
Wavelength 1 (numerator) : Wavelength that will measure t he numerator value of r atio.
Default is 260 nm.
Wavelength 2 (denominator) : Wavelength that will measure t he denominator valu e of
ratio. Default is 28 0nm.
Wavelength Referencing : Select whether measure or not Reference.
Factor : Factor that will be multiplied by Ratio.

5. Simple Kinetics
1) Test Mode Screen
At this mode, you ca n measure continuously absorbanc e variation at a wavele ngth during a
certain period of time.
2) Mode Setup
Wavelength : Input wavelength to measure. D efault Wavelength value is 6 00nm.
Total Scan Time : Input total scan time. Default dur ation
Interval Time : Input interval tim e to measure. But, Inter val time must lon ger than Total
Scan Time/241.
3) Measure
After Mode S etup, start measu ring with ME ASURE [F2].
You can see progressing r esult with absor bance as time variation.
After measur ing,MEAS URE [F2], GR APH [F3],
ABS/% [F4] butto n show up like the left s ide
gure.
